Webroot Spy Sweeper is the best anti-spyware you can buy (see the Consumer Search source for more information).
Still, no single program can detect and eradicate all spyware. It is best to use multiple programs, I recommend using Ad-aware and Spybot which are among the best (they are free).
Running these programs in Safe Mode (tap F8 after powering on the computer and choose to boot to Safe Mode) can be more effective.

Here's the combo of software I use.
I've been using this combo for over 2 years and not had one problem with any sort of malware.
Webroot Spy Sweeper $29.95 + $19.95 each year after
AdAware (free version or Pro) Pro is $40.00 but free is fine.
AVG Antivirus (free version)
I also have Spybot (free). I also have a couple of other programs that I run from time to time but they aren't free.
I also use:
Spyware Doctor - good program but needs 70M RAM after it boots up, and the boot up and shut down is quite slow. I simply don't allow it to boot up and I only run it on occasion. The programs above do a great job and I don't feel I have to run this unless I simply want to run everything I have in my arsenal to make sure my hard drive, etc. is clean.
Registry Mechanic - cleans out the registry. There are free ones at download.com - just search on "registry cleaners". I downloaded and tried one called Registry Distiller that seems to do a pretty good job of it.
Diskeeper - $29.95 automatically defrags your hard drive and keeps it cleaned up. You never have to remember to run the MS Defrag utility again. It also runs in the background and will give up the cpu to what you are doing and then will resume when you are done. Great program !!!

The best anti-spyware and anti-adware you can buy is common sense. To the best of my knowledge this is free but you could also take a course in security awareness.
If you have a good antivirus/firewall suite installed from a reliable vendor such as McAfee.. and you keep the program properly configured and allow it to automatically update, etc.. then you should be well protected provided you have good/safe browsing habits. (common sense).
If you are infected with spyware that your antivirus has not detected/removed you could download Spybot, which is free.
