I like Mel Borooks's "Young Frankenstein", "Blazing Saddles", "Spaceballs", and "Robin Hood: Men in Tights".
Woody Allen has also made some foney films, "Take the Money and Run" and "Sleeper".
That said, "Monty Python and the Holy Grail" is a true classic. I saw the musical "Monty Python's Spamalot" recently, it's based on the film and it's hilarious.

I love the Holy Grail, but some other comedies I found to be equally funny, though quite different are Bringing Up Baby with Cary Grant and Catherine Hepburn and Some Like it Hot with Marilyn Monroe, Jack Lemon, and Tony Curtis. Hilarious!

Nothing. Something else may be almost as funny. The Jerk, 40 Year Old Virgin and Some Like It Hot are all hilarious. The other Python movies are good for a few laughs, but nothing compares to this quotable classic. Bring out yer dead. Also if you have not seen A Fish Called Wanda I must agree with a previous poster, John Cleese and Kevin Kline make this film it is a must own.

The holy grail was fantastic but I don't think it was the greatest commedy of all time. It is in my top 15 though. It would probably rank in at 7 or 8 "She turned me into a newt!.....Well I got better"

in the area of slapstick commedy: history of the world part 1 or blazing saddles....actually, almost ANY mel brooks movie would be a good choice...
the greatest commedy, however, is "the big lebowski." its not a slapstick, but it is GREAT!!!
