One of the most popular 2 player games is "Lost Cities" by Reiner Knizia. This game is excellent for husband and wives and plays fast.
Another game that is great for 2 players but also is very fun with 3,4 or 5 players, too, is "Carcassonne".
My wife's favorite game to play with me is "Settlers of Catan Card Game". It takes about 1-1/2 hours to play and is a little more difficult to learn if you're not familiar with Euro-games or have never played the Settlers of Catan board game, but it is a lot of fun.
If you're not looking for a couples game and instead looking for something that 2 buddies can play, check out "Battlelore". It is strictly a 2-player game and is one of the best, most fun games ever made (in my humble opinion!)
For some great reviews on these games and also an online store from which to purchase them, check out www.funagain.com.

I assume you're NOT talking about "adult" games, with sex and drinking.
I'm a huge fan of many fo the games from Looney Labs. Their Icehouse/Treehouse games "system". Treehouse itself does basically work for two people but is more fun with three or four. But once you get three or more Treehouse sets (they're inexpensive), you can play MANY more games.
For instance, Looney Labs sells a little booklet called 3HOUSE that gives the rules for three games you can play with the pieces from 3 Treehouse sets. Black Ice is a code-breaking game with a nice mix of strategy, memory, and
dice-rolling luck. Martian chess, which requires a chessboard, is a chess variant with a funky twist. And lastly Homeworlds is deep full-information strategy game. The number of choices available in Homeworlds on each turn make the game highly replayable.
My favorite games using these pieces are Zendo and Volcano. For these you need five Treehouse sets. Additionally you'll also need something to use as "caps" and a board for Vocano or some type of colored marker (glass beads, othello pieces, etc) for Zendo. Of course LL also sells these other bits, but you can just as eaisly improvise with things around the house.
Outside of the Looney Labs universe, the only other game the wife and I play on a regular basis (other than regular old card games) is Carcassonne. We play with the tiles from the basic set and all the expansions, but use just the basic rules plus those for the Inns and Cathedrals exp.

I have been through billions and billions of board games but one that I have been dying to get my hands on for years now is one called Cards Against Humanity. It's not quite a "board" game because you only use cards, but it is loads of fun. You may have to order it online so search it up on Google, see if you are interested, and then order that baby up!

2 adults? most board games aren't fun unless there are more than 2 players. Cranium is fun, pictionary, pop culture trivia games are fun. The Scene It games are popular as well.

Depends a lot on what you like!
Lost Cities is a popular two player card game. In it, you try to build sets of cards (themed around exploring lost ruins) to get the best score. Its fast and easy to play.
Lots of details: http://www.boardgamegeek.com/game/50
If you enjoy party type games, Cranium Hoopla is the only one I know that also plays well with 2; all the players compete as a team against a timer, with activities like charades, pictionary and word games. Also great with big groups!

Want a challenging and tense game with an element of mystery? In Mr. Jack, one player tries to catch the infamous Jack the Ripper, while the other tries to make him escape. But he could be any one of several suspects!

You can play this one online as well at http://mrjack.biludi.de/
How about taking railroads across the country? Ticket to Ride is an easy and fun where you collect cards to get railroad routes; scoring more points for longer routes. But you have to be careful; an incomplete route counts against you, and your opponent can block your routes! Plays well with anywhere from 2 to 5 players.

Feel a bit more confrontational? Nexus Ops is a bit like Risk, but its very fast moving and exciting (a 2 player game probably takes about half an hour) with lots of interesting choices to make. It feels a lot like playing Starcraft as a board game.

There are tons of awesome games out there - if you can give any info on what themes you might like, or what sort of games you enjoy, I'm sure there are more specific recommendations to be made!

Sequence, it's kind of like Othello but you use cards to play too. You can play with two players or more.
I just gave you two fun boardgames to play.
