space shuttle is no longer true.

its valid, but there are now 2 (actually 4 but read on)

spaceshipone is the second and owned by a private company

it is US funded, designed and flown.
It has not been used as much as the shuttle, and neither are flown or in orbit at this moment.

It's version 2, spaceship 2 WILL be in air shortly on a daily basis (but it doesn't count yet)

Six shuttles have been built, five of which were spaceworthy
Columbia, Challenger, Discovery, Atlantis and Endeavour


Challenger was destroyed in 1986
Columbia was destroyed in 2003.

Endeavour will be used in oct 2007
atlantis will be used in dec 2007
endeavour will be used in 2008

The ISS is not US property, so even though there are Americans on board its not technically a US manned craft.
The only other US craft that enters "space" is the space shuttle, but there isn't one in orbit at this time.

So the answer is none.

If you mean right this minute; zero.

The international space station has americans on board right now, but it's not a US spacecraft, technically.

The space shuttle is the only truly american manned space craft in use at this time.
