I'm staying in Millbrae, CA and would like to take the kids to see Grace Cathedral, Lombard St and Fisherman's Wharf within 1 day. What is the best way to organise this in terms of transport.

Suppose you don't have a car, take public transit.

BART from Millbrae to Powell station, 30min
Cable car to Fishman's wharf 30min, you will pass Lombard Street. You can stop over, but you have to purchase another ticket unless you have day pass.
Street car to Ferry building, 15min.
Another cable car to Grace cathedral( nob hill) 15min
Walking down to Union Sq. 15min
Take BART back, 30min.

So you have plenty of time to see SF.
