Yep, sure can. Don't even need a router, but a switch at minimum.
Allow sharing for both computers/drives and copy away.

you should be able to buy a crossover cable- it is a special cable with the send/receive wires reversed which eliminates the need for a hub/router.
you can get these at any computer type store, office max,office depot, compusa, etc, should be about $7 or so.
just plug one end to computer #1, the other to computer #2, enable file sharing on both machines, and you should be able to see the files you wan to transfer, just copy and paste...
