Didn't they actually have two games last Monday night during opening week?
The reason for it, traditionally, is that it would be hard to get great ratings for two games when the second game would begin when the whole East Coast has gone to bed. The Monday night game was intended to be the feature matchup of the week, a game that could be watched even by those who couldn't watch the Sunday games. Of course, that has changed now that the "feature" game has moved to Sunday nights and MNF has lost much of its luster by being on cable and having some second-rate games.

Ratings and players union = 1 game. It's an easy way to capture the TV market for huge dollars as Monday has little else to compete with. The players/coaches don't like Monday's because it reduces recovery and practice time for the following week. If they had more than one game, a lot of the elite teams (always chosen for ratings) would face this hardship significantly more times than others and be at a competitive disadvantage.
