A person of substance has the power and intellect but the person with the right material has the actual skill or knowledge required to do the job. Having substance may make you mature and wise but you still need to be thorough in doing a job thus you need the innate knowledge or innate skills to be able to have the right material to accomplish the task. Substance is 'power' but right is 'apt'.

A person of substance has depth. They have a quality that is difficult to define other than to say they are not superficial. It usually implies a moral, ethical, or spiritual depth that is normally uncommon. A person with the right material refers more to the physical, mental, & technical/educational aspects of a person. A good example is the apollo flight crew had to have the "right stuff". A person with the right material does not mean he is a person of substance, & many a person of substance are not educated, so the 2 do not always go hand in hand.

It's the depth, the insight, the knowledge that could be called a person's substance. His rich and profound quality that produces a meaningful well-being in him where he stands in reverse of a shallow person.
A person with the right material is considered a skilled one who has the right aptitude to get something finely done. It's a talent for something specific. A fit-for-the-job kind of thing.
If a person of substance has the right material; then he is an excellent breed.

Maybe it's just your sentence structure, but 'of' substance denotes that the person themselves has in their being that which makes them valued... substance being the 'under' structure that is at their core.
A person 'with' the right material denotes someone who has gained that quality that is valued. It is not 'of' them, but an appendage, and add on. 'Right material' in itself is something constructed not natural or pure.
A person of substance and a person with the right material might both get the job done, but a person of substance gets it done because they have to; it's in their DNA.
