I think one can get a good general impression of SOME of a person's character/characteristics at first meeting but, from experience, I know that one can only truly find out what a person is like over time (how they respond to different types of people, events, situations, etc.).

Not necessarily. I'm a lousy judge of character. I tend to think everyone is lovely, and only find out the Awful Truth much later.
On the other hand, I do find it hard to get past my initial response to someone: the way I behave towards them, I mean. If I start off patronizing, or cringing, or jocular, or flaky and helpless, I keep doing it. long past the time when that was appropriate, if it ever was.

Sometimes. I know a person may be, and may not be how they appear...a series of impressions makes a better lasting impression.

Not always. I thought one of my girlfriends work colleagues was a bit of a creep when I first met him. How wrong could I be. He is a true gentleman, with a great outlook on life. I still don't know what I was thinking.
I give everyone a chance.

In my experience i must say no. On a few occasions i have disliked someone by simply observing them from a distance. I have thought the person to be so not on my planet. when i have made their aqquaintance i have found them to be great people. I would definately say you can`t judge a book by looking at its cover.
