Only when you can walk away and KNOW that there was nothing more that could be done to save it.
After you've tried mediation and/or counseling. Only after every avenue has been tried, after every effort to save it has failed, after all the anger, disappointment, and issues have been resolved, because then you can go on to your new life knowing that it truly was over.
It leaves you with less baggage to carry into any new relationship, and you can close the door on your marriage, not with a resounding bang, but a soft click.

When you don't care about each other anymore. When you don't trust your partner on what he/she is doing. When the reason why you're staying together is just for the sake of your children. I know that the divorce rate is higher but if you don't feel anything for your partner anymore, why suffer? If it will make both of you happy being in separate ways then be that way. After all there's not a thing such as a perfect marriage.

When you are waiting for someone else,
and nothing the one you are with can do to compare,
I cannot say for certain it has to be over, but, i know in my heart it is, i think you just reach that point and then you accept it, even if you know no one else is waiting.
best of luck...

when the two of you really thinks that its not really working and theres no love for each other anymore... its a tragedy for anyone... if you can, and while you still can, try to salvage your marriage coz it might still be worth it... try counselling and try to really examine yourselves if there is something that you feel for your partner.
