The main difference is that the NFL has vastly superior athletes than the CFL. And don't believe this "linemen in the CFL are faster". The NFL is full of 300 pound guys who can run the 40 yard dash in under 5 seconds, which is amazing for that size. They want guys with both size and speed, so the offensive lineman have to be quick to take on speed rushers like Dwight Freeney, Julius Peppers, and Michael Strahan. The NFL is a faster game, the fact that the CFL has only three downs does not make its players faster. The best college athletes, meaning the biggest and fastest stay in the U.S. and play in the NFL, those who don't make it, the small, slow ones end up in Canada. So do the weark-armed quarterbacks, like Ricky Ray, who is great north of the border, but couldn't even stick as a third-stringer two years ago with the Jets.

another difference is that nfl rules allow
the ball to be placed as near the goal line as possible without being called a touchdown. the cfl allows the ball to be placed only at the one yard line.
another difference is the line of scrimage. when the ball is placed on the field, the defence can line up no closer than 1 yard away. the nfl allows the defense to line up as close to the ball as they want, as long as they dont break the plane of the line of scrimage (which is probably set by the nose of the football prior to it being jacked up by the centre. lol :)
another difference is that in the nfl you can only score a sinlge point by a convert.
the cfl allows single points to be scored in other ways than a convert.

Colin covered it pretty well, except the field is also wider, so the linemen are called on to cover more ground so they are often faster than NFL linemen (and smaller). There are a few 300+ guys in the league but 280 and fast will get you the job.

the CFL plays in a stadium half the length/width as the NFL
and the also are governed by college rules
