Not that any of these people would do such a thing, but it's probably a good idea to avoid sending anonymous folks over the internet your entire manuscript.

Excerpts, send them excerpts.

I am an editor for a subsidy publishing house (sadly, private because we print mostly religious or geneology tomes) and I can tell you from the manuscripts we get that the novel must fit certain genre, certain length, and be marketable. The first two are easy to do if you look in the Writer's Market (get it at the library) and edit carefully. The third one changes all the time so what doesn't sell today will tomorrow, so keep sending the novel out. Rowling was rejected (as was Stephen King) because of they didn't think it would sell, so you have to keep trying.
Good luck.

I'm sort of a real bookworm, but I'm also really picky about what I read. So I'd be more then happy to read your manuscript and tell you what I think. And ask for the rejection letters, don't give up. Lot of other successful authors were rejected, but in the long run, their books sold like hot bread. Here's my e-mail:

Do you know how many publishers rejected J.K. Rowling before 'Harry Potter' was picked up? Or how many times Stephen King received rejection letters before he published his first novel?

You cannot allow one (or several) 'no' letters dissuade you from trying again.
