Maybe he looked so sweet and endearing saying "I do" that she didn't just smile -- she laughed, out of sheer joy and happiness.
Or maybe she was laughing out of relief -- i.e. she never thought this day would come, so hearing him say "I do" was both joyful and relieving at the same time.
I dunno. I haven't really analyzed this particular song, although I've heard it enough to be familiar with it. :)
