The Union Pacific 4-6-6-4 Challenger locomotive. The largest ever built was the Union Pacific 4-8-8-4 Big Boy.

The key here is operating. It is my understanding that the one rebuilt to pull the centennial train in 1976 was the biggest example ever built and is still running.
