It totally depens on what KIND of business you're doing. Anything with food would require licensing from the health department in your city. Animals are another department. Wholesaling is different than retail, and if you're not selling any 'goods' but provide a service instead you may need special licensing (HVAC, electrician, etc.) from whoever certifies that sort of thing.

Standard city licenses would be able to be researched with the City Clerk in your phone book, and they can also point you in the right direction regarding getting your business registered with the state. If this is to be done from home but you don't have customers come to you, a mobile sellers permit may be needed (they're pretty cheap). If you have a store, then that's completely different.

When you decide what you want to do, try www.google.com searches with that business and see what comes up. Also contact your local SBA or SCORE office, they can walk you through all the processes.

You'd best start with a passion, be it coin collecting or water purification or whatever. Once you're confident enough about what you're doing (meaning also that you are able to make a profit - eg, even by going to garage sales, buying some item for a buck and selling it for three...) then you're on your way: the drive to make a profit will take care of a number of things. Then when it comes time for licenses you may look for more specifics through the references at the library, the internet, or perhaps the services of a lawyer.

I suggest you find the best restaurant in your region and get a job with them for a minimum of 1 year. Learn everything you can about running a restaurant while getting paid. Then, if you are still convinced this is something you want to do, begin by doing some research about the local market and finding a good location.
