Governor White, the grandfather of the first child of English descent born into the new world (Virginia Dare) left his colony. He wished to plead with the crown because due to war (with Spain) Britain was not sending enough food (colonists hadn't yet been able to successfully plant crops) medicine and weapons. Pirates and privateers loyal to Spain were a great threat to the under protected colony.
When Gov White left the situation was critical. Due to the war he was delayed for a long period but did return with the needed supplies, finding the colony deserted but with the word Croaoan carved on a tree.
Gov White knew of a friendly tribe on a nearby island called Croatoan but due to weather the ship's captain would not sail to the island.
It would be some time before Britain planted a new colony and by that time most of the Croatoan had moved on and there was no evidence in their involvement in the colony.
Decades later Croatoan were found in what is now Kentucky, many of these native Americans had English names, blue eyes and fair complexions.
The tribes lack of written records means that there will never be an exact by the book answer, but it is reasonable to assume that colonists joined the tribe and assimilated.

They moved further south to be with the Croatoan tribe then went inland. The Croatoan's were around the Hatteras area and Roanoke Island is north of Cape Hatteras. Going north would have brought them into further contact with tribes hostile to them. But Manteo's mother's people, the Croatoan would have been friendlier to them.

Knight is right. The accepted theory is they couldn't make it without supplies from England so they joined the Croatoan and carved the name of the tribe on the tree.

When Lewis and Clark went into the West they found the Platte head or the Indians of the Platte river, many of them with blue eyes and blond hair. It was never explained or investigated as to way.
There were also stories of the Kentuckians that moved into the Appalachians and were settling the areas around the Orleans areas and Kentucky, the same oddities, Indians with light skin, blue eyes and blond hair.
It's all lost to time and history now.

I cant remember the name of the program but there was some genetic testing done on some folks that claim to be decedents of the folks at Roanoke. Turns out there was some truth to their claims. The program shows a genetic linkage between to descendants of common English ancestry. And the one in the US is from one of the settlers that left England to come to Roanoke. Pretty good proof that at least some of the settlers melted into the native population.

Nobody really knows what happened to Roanake. When Roanake was found again then all the supplies were there just like they had just left. There was a X on the tree and historians have many theories about this.
