To wink is to close and open either one or both eyelids with a rapid motion; to blink suggests a sleepy, dazed, or dazzled condition in which it is difficult to focus the eyes or see clearly. A wink is a form of semi-formal communication, which indicates shared, unspoken knowledge.
A "naughty wink" can silently indicate a shared secret, such as if a salesperson gives a customer a brochure and says, "Here you go; it's free". Infrequently, it may also mean "got it" or "yes, I understand".
In Western cultures, women may wink to men they are interested in dating, but this has grown out of fashion, though still used occasionally. Winking is also done by men to women.
In Latin American cultures, winking is also a romantic or sexual invitation. In Nigeria, winking is a signal for children to leave the room. Many Asians, especially Chinese and Indian women, consider winking to be rude.
Not all humans are able to wink voluntarily, and some can only wink one (usually the non-dominant) eye but not the other.

I'd go with: he is hitting on you...or he is flirting with you. Depends too, do you know the guy? Cause I have a guy that when he passes by my office he winks at me or gives me smooches but I know he is just kidding around cause he makes funny faces and I've know the guy for 7 yrs so I know his character more and what not. If he is a total stranger just winking at you, then that means he is just trying to flirt with you. Found you attractive and decided to wink. Some men are brave or just cocky! LOL

Winking is an expression of a made up agreement. So, it's implying that you both share a made up idea.

Depends on situation.
Just being a prankster(just for fun) or may be a message to let you know he likes you or even he is playing a prank with some one and he thought you observed that too and he signed you to keep watching the fun.

Several options open themselves up.
1) He's interested in you, and trying to get you to notice his nonverbal advances.
2) He has a nervous tic.
3) Something in his eye.
4) Getting ready to have a seizure.
5) Is atttempting to share some in-joke with you.
6) Something is going on in the conversation, and he wants you to play along.
Either way, it's all a matter of context.
