I'm not sure about the top 5, but the most valuable card ever is the 1909 Honus Wagner card. Back in those days baseball cards were used as promotional items in packs of cigarettes. Wagner didn't want to promote smoking though, so he made the cigarette company stop using his card. Because of that, this card is extremely rare, and that coupled with the fact that Wagner was a great player makes this the most valuable card. I think one sold at an auction a couple of months ago for over a million dollars.

EDIT: Wow, it's even more valuable than I thought, 2.8 million!

I know for a fact the #1 sought after (and most expensive) card is the T206 Honus Wagner card. It's been sold for 2.5 million.

In no particular order, I believe another the other 4 cards are:

1952 Mickey Mantle rookie card.
1914 Babe Ruth rookie card.
Joe Jackson (I don't know the year or brand)
Ty Cobb (I don't know the year or brand)
