No there is not such thing as a Tune Up any more. That is just a marketing Buzz Word for maintenance. Today's car computer tunes the engine like 100 times a sec or so.
A tune up is the old days of readjusting fuel metering systems (carburetors), ignition (points and trimming), idle, spark plug cleaning / gap check and such. Cars back then were preset by factory "Tuned" to an average range of conditions, so when you drove them in winter or summer, high altitude and sea level the settings changed and cars ran rough, Therefore........ you had to "Tune Up" the car for optimal performance. Tune it for the weather conditions.
With the advent of Computers that work is completely gone for good.
Changing spark plugs, check the wipper blades, water level, air filter, oil and filter change, tire pressure check, etc.... is not a Tune Up, that is just maintenance.

A transmission flush focuses on the transmission alone and generally only the fluid in it are changed and checked. A tune up is changing the plugs, wires, air filter, and the oil at this time most fluids should be checked not necessarily changed

No. The tune up is what they do to make your motor run more smoothly. Usually, they replace parts, and they check all the computer stuff.
A transmission flush is to make the car shift gears better. The transmission has a bunch of special liquid in it to make everything run well, and that liquid gets all kinds of metal shavings and other junk in it. The transmission flush is just what it sounds like - they flush all that junk out of there, and replace clean, new fluid so it works better.

Depends on the shop, most likely if you're looking for the most fluids changed go with the tune up. don't be afraid to ask.

tune-up is usually changing the spark plugs, but some places will also check & change your transmission fluid.

No a tune up is when you get your spark plugs replaced.
Transmission fluid is what your transmission needs to be lubricated. I should probably by your owner's manual recommendation.
