No. You should NOT copyright it. It is considered unprofessional. It shows an agent or publisher you don't trust them. Trust me - NO legitimate publisher or agent is interested in stealing your work. It is insulting to them when they get a manuscript that is copyrighted. It shows you are an amateur and haven't done your homework regarding publishing procedures.

You already own the rights and as long as you keep it off the internet, you won't have a problem. If and when you sell the book, the publisher's contract will include a clause stating they will copyright for you. That is standard. All authors know this. It is in the contract automatically.

Never send a manuscript to a publisher or agent unsolicited!!!!!! You only query agents online who accept queries online. Not many do yet. What you will be doing is sending a query letter and a synopsis first according to the particular publisher's submission guidelines. Not the actual manuscript.

Have you had it edited? You should do that before you query. If an agent is interested in your query, they will write to you and request a full or partial manuscript. Most will request it sent via snail mail. They won't be expected to print it out to read it.

Put it away for a couple of months and don't even think about it. Then, get it out and read it. Don't stop to rewrite anything until you've read it again--cover to cover.
Persiphone was right about editing, but you should have some good readers review it first--there's no point in editing if you're going to find that some major surgery is needed.
By 'good readers' I mean people that are familiar with this type of work (you didn't mention if it is fiction or nonfiction), and can give you brutally honest feedback. Trust me, it ain't easy to find such people. When you do, cherish them. They are a writer's best resource.

Self publish, publish on Amazon or find a reputable agent. Would suggest not sending your ms to anyone online.
