It all depends on how she says it. I know many women that have said no but meant yes. However, I think there is a very fine line. If a woman says "no" and means it, you should definitely stop. I think it is pretty obvious to know the difference though.
What is really weird is I just saw this T-shirt at www.recklessts.com/noyes.html that says "No sometimes means yes". Pretty funny stuff.

Depends on the woman (sometimes there's the whole "struggle" fantasy thing, but generally only if she knows her partner VERY well).
Always safest to assume that she really means "No" and let her berate you afterwards for spoiling her game than carry on and end up in a situation that's unpleasant for everyone. If you're together for long enough, you'll certainly know.
In short... err on the side of caution.
Edit - I'm assuming that those thumbs down are from rapists?

Yes If I say no I mean it. Too many men look on no as somekind of challenge to prove she really meant yes. If you are talking sexual context NO means NO.....and if you continue then need I say more.

It varies from woman to woman, but if a woman says no, you'd be well advised to take it as a no, whether you think she really means yes or not.
