If you are using a dedicated leased line, it only runs between two points. Think of it as a LONG ethernet cable. What you connect on each end is up to you. Sometimes you do use a router on each end and sometimes only on ONE end.. all depends on the network you are building and how you want them to communicate. This is A REAL network line and not a VPN (virtual private network) You don't need virtual when you have real!

A VPN creates a tunnel within the internet lines to make a private network.. A dedicated line isn't open to anyone else so there is no need to tunnel it at all. It is just a long wire between two places.

If you are talking about two computers that are remote to one another, should be able to do it. There will be a modem of some type at each end that will provide connectivity. Use a VPN connection for security to make the connection, or the remote access connection from one to the other. Remote access will only be one way data in some cases.
