If you're looking for less well-known, be aware that one reason plays like Henry VI are less well known are because they're not as good.

Hotspur and his wife have a good scene in Henry IV part I.
Brutus and Portia in Julius Caesar.
Antony and Cleopatra in the obvious play.
Coriolanus and his wife have a good scene early on.
You might also go with a female Mephistopholes and Faustus in the play by Marlowe.

In Twelfth Night, Act II Scene IV, there's a bit of a scene between Viola and the Duke -- she loves the Duke, but she's dressed up as a man to work for him, and now the Duke is sending her to Olivia, the woman he loves, to woo her for him. It's so totally romantic, and also a bit sad, since he's so obsessed with being in love that he can't recognize what's right in front of him. Anyway, it's a really good scene, and also not very well-known.

The first act in A Midsummer's Night Dream. It is a fairly well-known comedy. Most have heard of it, but they don't know what it's entirely about, so it could fit your needs. The first act has an intense moment between Hermia and Demetrius. It sets up the conflict for the show, so it's fairly dramatic.
