Dude, if you are talking about cash then I'm thinking money laundering. And the banks will assume the same thing.
So, best thing is to split you USD 500k into at least 50 bundles of max. USD 10k and visit at least 50 different banks or money changers .... you'll have a busy day.
If you have the money legit in your bank, then simply ask your bank to move the money from your USD account to your (new) JPY account.

According to this, "There is no limit on the total amount of money and other means of payment that may be brought into or taken out of Japan. However, if you transport money and other means of payment exceeding JPY1,000,000 or its equivalent or precious metal exceeding 1KG in total weight, you are required to submit a report to Customs. 'Money and other means of payment' includes Japanese or foreign currencies, checks (include traveler's check), promissory notes and securities."

It's not possible unless you're trading currency in which case you go through a broker and pay a fee.
If you try to do it all at once, the bank will think you're either trying to launder the money or you got it through illegal means. This means trying to prove to them that you got the money legally.
If you try to break it up into smaller amounts and take it to bank after bank trying to get it changed that way, you're going to be at it all day.
So there's no real way to do this without a hassle.

No hassle free way if you don't want a paper trail. Break it up into units under $10000 US and take it to different locations-"smurfing".
