In that situation there are two ways you can succeed.
1) Have six or so friends to occupy the others and pick up ALL of them between you.
If not, the chances of getting blocked are way too high to bother trying so...
2) Get the chick alone start making out with her, take her out back to the Ladies toilet and seal the deal in there. No leaving the club, no blocking. Yes. This is a useful option in many other situations also.

I have only done that once before. And in that particular situation, it was because I knew my friend was totally drunk and didn't know what she was doing, and that the next day she would have regretted going home with that particular guy (he was old and nasty). I managed to get her outta there, and she thanked me the next day.
Anyway, I think overall the reason women do this is just to protect our friends. Nowadays a woman going home with a stranger to sleep with him is still considered a bad thing (she's a slut) whereas with a guy, taking home a strange girl to to sleep with her is viewed as a good thing (he's a stud or a player). So we're really just protecting our drunk friend from making a potentially damaging choice that she might regret when she's sober. We're not trying to hurt our friends by doing this, we're looking out for them. More often than not, women thank each other for this. It's not being a "killjoy", it's being protective.
All that being said, I can understand how you'd be frustrated. Especially after buying her all those free drinks. Just try to pick girls that are on their own (not that that ever happens) or maybe get your buddies to flirt with the other girls. Maybe if her friends are distracted, you can sneak her out.

The friend who blocked you is concerned for the girl who would've otherwise left the bar/club with you. The woman you want to sleep with probably doesn't know you, the woman who blocked your "play" probably doesn't know you, so going home with a stranger isn't a *smart* thing to do in anyone's book but the player's (in this case, yours).
It's a known fact that when women go out on a girls' night out, 99% of the time no one is expecting to get laid by some guy the group meets at a bar. This isn't a "player and her wingwomen" situation, this is chicks going out to dance, sometimes consume alcohol, sometimes flirt, and go home. If you like the chick, give her your name and phone number, wait a few days, and see if she calls. If you don't like her enough to do *that*, try again to find someone who's after the same thing you are, or just write it off as a loss, go home, and amuse yourself.

Yeah...what the first poster said. One of my friends was a virgin all through college, and she wanted to stay that way, but when she would have some drinks she'd fall all over the nearest guy. But always before we'd go out she'd make us all swear not to let her go home with anybody.

Smile. Women take care of their own. You really should find out more about the people you are trying to sleep with, before sleeping with them. This friend might of heard something you have been trying to hide?..

We look out for each other. If one of us says "don't let me go home with a guy!" then we honor that. If, on the other hand one of us says "I need to get laid tonight" then we hand her a rubber and tell her to have fun.

Because we are considered sluts if we meet some random guy and sleep with him but you are considered a stud. Our friends are looking after us, and you're friends are looking after you.

well, i hate to tell you this, but the girl might have said something to her friend like "i don't wanna go home with him but i don't want to hurt his feelings so can you do something?" or she saw her friend was really drunk and possibly not able to make very good decisions.
