Not all "superheroes" wear the same sort of costume; Prince Namor the Submariner usually just wears a pair of swim trunks and Speed Centaur wore no costume or clothing whatsoever (he was a centaur and had no need for clothes). Some "superheroes" like the Spirit and the Clock (who is arguably the first original "superhero" created for comic books) wore ordinary street clothing and a simple mask, and other characters, like Mr. Risk and Master Key, even eschewed masks.

If by "wear spandex" you are asking why most of the cape and cowl set wear distinctive costumes, there are several theories for the convention, but the theory I favor is that the costumes can all be traced back to the designs of the clothing worn by acrobats in circuses. Obviously, the costumes are eye-catching and those in the know can recognize a particular character at a glance, both factors lending themselves to cover designs. Another theory holds that the costumes were used because much early comic book artwork was so primative that it was hard to tell characters apart and the costumes simplified that task. Some costumes even have components which help explain the impossible (like Iron Man being nearly invulnerable because he wears armor, or Hawkman being able to fly because he wears a pair of wings).

The convention of tight fitting costumes is actually older than the development of artificial fibers, like spandex, and I somtimes wonder what fabric costumes were made from back in the golden age of comic books. I think knitted silk and wool are the best candidates for the mystery fabrics worn by mystery men.

The old ones used to but now hardly any of them do. Mostly just Superman and Spiderman. Most of them where some sort of protected armor of some kind, even Batman has Kevlar instead of just the tights as he once had.

The reason that a lot of them did have spandex was because you dont want to trip all over yourself when your chasing down a bad guy jumping from roff top to roof top.

The reson that is given for most heroes wereing a skin tight costume is so they will not get caught on it. The idea of a costume is for ease of identification and protecting the secret idenity. Spandex is not the material of most of the costumes many of them are from newer high tech fibers or even alien materials.

I guess it's either to make them sexy, or because it's more flexible to wear so they can do acrobatics better.
