An excellent question, and deserving of the star that I will bestow upon it.

My personal choice is that of One Flew Over the Cuckoo's Nest.

Without spoiling it for those who have yet to see this cinematic masterpiece, the catharsis of the central characters is both deeply saddening and yet what immediately follows is one of the most uplifting pieces of celluloid, that no matter how often I see this film, it brings a lump to my throat and tears to my eyes.

Also there is a bittersweet moment of irony in Christopher Lloyd's character that only lends more gravitas to this section of the film.

In fact, I will have to go and wipe my eyes thinking about it.

Definitely The Usual Suspects gets my vote, or the 70s remake of Invasion Of The Body Snatchers. When Donald Sutherland turns to the camera and lets out that wail, it is chilling.

Of course, Speilberg's ending to E.T. is also a true classic.

The ending of 'One flew over the cuckoo's nest' was excellent.
Indeed the whole film is a masterpiece. It's both the funniest and saddest film I've seen and the actress who played Matron was brilliant.I could watch it a thousand times and never tire of it.

The Sixth Sense (which happens to be my #1 favorite movie) has the best ending in movie history.

Without a doubt it has to be Usual Suspects - brilliant! Although Fight Club is pretty damned fine too.. . . . hang on a minute! What about Sixth Sense? Or Devil's Advocate?
I don't know!
You decide!

Either......
John Carpenter's 'The Thing', where McReady And Childs are the only ones left and neither knows if the other is infected

Or

David Cronenbergs 'Dead Ringers', where Bev seems ready to return to his life but then returns to the body of his twin

Or

George Romero's 'Night of the living Dead', (won't spoil it for anyone who hasn't seen it)
