I have a xps410 with a 375 psu and the graphic card I would like to get is a e8800gts that requires 400 psu. Will this make a big differences in the function of this graphic card???

The difference is between able to function properly and not functioning at all. It is a big difference.

If your PSU does not meet the requirement, it may not be able to supply enough power to your graphic card. Once you start loading your PC by playing 3D games for example, the PSU cannot supply enough power to your system, you'll start facing problems such as the PC hang and stop responding, or it will shut down completely.

You may also risk damaging your PSU, and when a PSU damage, there's always a risk of damaging your hardware.

You'll need a pretty powerful PSU to run a high-end card like that. I'd say go for something around 600W to be safe. It's not that you'll see better graphics with a more powerful PSU - just you'll have less chance of frying your shiny new 8800 :)
