Moe, Curley and Shemp were brothers. The others were unrelated.
The Three Stooges were commonly known by their first names: 'Moe, Larry, & Curly', and 'Moe, Larry, & Shemp', among other lineups. The act originally featured Moe Howard (born Harry Moses Horwitz), brother Shemp Howard (born Samuel Horwitz), and longtime friend Larry Fine (born Louis Feinberg). Shemp was later replaced by brother Curly Howard (born Jerome Lester Horwitz) in 1932. It was common for Jewish perfomers in Vaudeville to Anglicize their last names in order to appeal to a wider audience; Horwitz became Howard and Feinberg became Fine.
When Curly suffered a debilitating stroke in 1946, Shemp rejoined the act. After Shemp's death in 1955, he was replaced by baldheaded sissy comedian Joe Besser, and eventually by Joe "Curly-Joe" DeRita (born Joseph Wardell).
After Larry's death, Emil Sitka, a longtime actor in Stooge comedies, was contracted to replace Larry, but no film was ever made with him in the role, although publicity photographs exist of him with his hair combed like Larry's posing with Moe and Curly-Joe prior to Moe's death. Larry's death marked the end of the act.

There are web sites that tell the complete story, but the others that I can think of were Curley Joe , Joe and Shemp. Moe, Curly and Shemp were all brothers by the last name of Howard. Larry was Larry Fine. There was Joe Besser and Curly Joe DeRita. The original act was called Howard, Howard and Fine.

Shemp Howard was the first to take Curly's spot. He was the oldest brother of the Howards (Shemp, Moe, Curly) Those three were real life brothers (The Horowitz'). Anyway, then came Joe Besser, and then Curly-Joe (Joe DeRita). Those last two were of no relation.

The three that were related were the Howard brothers: Moe, Shemp and Curly. No other members were related.
The original 3 were Moe Howard, Shemp Howard and Larry Fine. They started in vaudeville working under comedian Ted Healy, who was a boyhood friend of Moe's.
The trio split from Healy after a pay dispute, Healy threatened to blow up the theaters they performed at, causing Shemp to leave the group out of fear. The boys worked out their differences with Healy and Moe's baby brother, Jerome "Curly" Howard joined the group. After a few years of making movies at MGM, the trio split from Healy permanently after Columbia offered them a contract to make 2 reelers.
In 1934, Moe Howard, Larry Fine and Curly Howard started making their short films for Columbia....they would continue until 1947 when Curly suffered a stroke. Still under contract, Shemp Howard returned to the trio and picked up where Curly left off, Curly died in 1952. From 1947 until 1955, Shemp was the third stooge...in 1955, Shemp died from a massive heart attack.
Moe and Larry considered playing as a duo, but Columbia hired 3 Stooges and they needed a replacement. Most of their movies in 1955-56 were remakes of their Shemp shorts using existing footage of Shemp. They wanted Joe DeRita, but he was under contract elsewhere, so they settled on comedian Joe Besser as third stooge. Besser was a stooge from 1957 to 1958. Besser left for personal reasons and DeRita was available.
They felt that Joe DeRita resembled Curly, and they already had a Joe...so they named him Curly-Joe. From 1958 until 1970, Curly-Joe was the third stooge.
Larry Fine suffered a stroke in 1970, putting the act on hold. Longtime stooge co-star, Emil Sitka was brought in to replace Larry....nothing became of the pairing, Moe's health became frail. Larry died in January of 1975, Moe followed him in May 1975, putting an end to the act.
Healy was beaten to death in 1937, Besser died in 1988, DeRita in 1993 and Sitka in 1998.
