Nothing at all. He certainly can run and even be elected vice president, even if he has already served two terms as president.
Let's say that someone who has already served two terms as president (such as G.W. Bush) is elected vice president. Let's also say the president resigned, died, or became incapacitated while in office. In this case, the vice president would not become president because he would be ineligible to serve. In this case, the next eligible person in the line of succession would become president: the Speaker of the House (currently, Nancy Pelosi).
It's not required for all individuals in the line of succession to be eligible to become president. Carlos Gutierrez is currently Secretary of Commerce, and Elaine Chao is currently Secretary of Labor. Both positions are ordinarily in the line of succession (10th and 11th in line). But both Gutierrez and Chao are ineligible to serve as president because neither is a natural born citizen of the United States (Gutierrez was born a citizen of Cuba, and Chao was born a citizen of Taiwan).

Vice president isn't an elected office. I don't think there is anything stopping a president from being a vice president, but I don't think any would want the job!

The US Constitution. It limits a person to serve two terms as President. If Bill Clinton were VP, he would be first in line to be President if something happened to the President. But he cannot serve more than two terms.

Because according to our Constitution and amendments the president can only serve 2 terms as president and if he were vice president there is the possibility that he might be thrust into that job.
