I think the best way is to go by train.
There's a train going several times daily from Madrid (station Madrid-Atocha Cercanias) to Murcia (station Murcia del Carmen). Traveling time is just over 3 hrs, or if you stop at more stations in between some more (up till over 5 hours or so).
Also, the train from Murcia ( station Murcia del Carmen) to Barcelona (station Barcelona Sants) is running daily and takes you there in about 6,5 hours (or more, also here depending on the amount of stops you'd make in between).

note: salida means time of departure
llegada means time of arrival

And Tanju is right, Barcelona is a great city, with lots of wonderful things to see and enjoy!

I have not been Murcia, but Barcelona is Excellent ...
You must visit Gaudi Gardens and L' Sagrada Church is gorgeous, You must visit them and please say My Greetings ...
