If you had SPOKEN the question, it would be fair for you to say, the answer is the letter "T". But if that's how you mean the question, the way it must be written is this:
What's the first letter in "the English alphabet"?
Since you did not include the quotes, the rules say you are NOT referring to that specific expression but to the alphabet itself.
So the answer to the question you TYPED is "A".
