I don't know about a 6-8 obgyn test, but I do know that you can have her gynocologist give a pelvic exam and a pap smear. The pelvic exam will tell the doctor that whether or not she's had sex(b/c your daughter's hymen will be broken), and if she has had sex the pap smear will test for some STIs and other problems that can arise from having unprotected sex.
The most important thing is that since she's 16, she's old enough to consent, which means that nothing can legally happen to the person she allegedly had sex with.
I wouldn't do any of this, though, without her agreeing to it. If you're right, you'll lose her forever because she'll never want to talk to you again. If you're wrong, she's gonna get mad at you for invading her privacy and not trusting her. You have to talk to her, that's the most important thing. She may never talk to you again, it could kill all her self esteem that you don't trust her.

A broken hyman is not proof positive that your daughter had sex. Don't rely on that one way or the other. Unless the gynecological exam occurred within hours of intercourse (or unless she got pregnant), there is no definitive way to determine if she had intercourse.

I would strongly advise against attempting to test her in any way. If you're concerned about this, then you should ask her in a non-threatening way to tell you what happended. If you have a good relationship with her, and she trusts you to be mature and deal with whatever the answer is in an adult manner, you'll get the truth. If she has any reason to doubt that she can count on your support, if she gives you an answer you don't like, then you probably won't get the truth.

There MAY be a test to determine if your daughter is a virgin. That is the closest I think you will ever come to what you want.
