Are you looking for a simple opinion or a recommendation? Are you a couple...a family? Between San Francisco, L.A., San Diego, Lake Tahoe, Carmel, Napa/Sonoma...., the state is too big and too varied to answer such a general question.

I love San Diego and the surrounding area...Disney Land is also fun, LA and Beverly Hills is also a nice day trip its not that far of a drive from San Diego to LA just up the 5 freeway...I have been to California twice cant wait to get back..

Wow, tough question. Big state and no location/timing guidelines...
I'd go with Yosemite. It's a must while in CA at least once.
