well, in my wife's case she usually gets giggly and won't look me in the eyes. You can also tell by how they act, if they're uncomfortable. Another way to tell is if they try to change the subject. There's more but I'm probably boring you :)

There are many ways. Here are a small handful:
If they look up and to the left, they're conjuring up a story.
If they put a barrier between them and their "accuser," they feel uncomfortable and are likely lying (even something small like a set of keys).
Their voice changes pitch due to stress -- it becomes more high-pitched.
Their body language doesn't match their speech.
Their story has inconsistencies.
Their story is all "good," with no adversity or downside.
Check out a book called "Never be lied to Again" by David Lieberman (a former detective). It's a quick & interesting read.
