The first formal, provable write-up about prime numbers is in Euclid's Elements a.k.a. Stoikheia in Ancient Greek (approx. 300 BC). Mostly in Book 7.
Factorisation is explained. Somewhere in the Elements, Euclid gives an algorithm to find the greatest common divisor of two numbers. If the two numbers are relatively prime (meaning they do not share a prime factor), then the algorithm finds GCD = 1. The description of the algorithm supports the idea that primes and prime factorisation was not only already known, but also already used at the time.

Because of the way the Egyptians calculated fractions, it is thought that they knew about prime numbers (at least in principle) even before the Greeks. Their method of calculating fractions implies that they knew about prime factorisation (but we can't prove it)

An archeologist (de Braucourt) found a bone portion on which were carved notches corresponding to four prime numbers (11, 13, 17, 19). If these notches truly represent an identification of prime numbers, that means that primes were known 20,000 years ago. However, the best interpretations indicate that the primes are used in additions, not factorisation.

From Wikipedia, I found the discovery is at least as early as the Greek mathematician Euclid who knew and wrote about it in 300 BC.

Prehistoric ( cave man ) did.
animal bones with prime numbers of tool scratches in distinct groupings were found suggesting ancient prehistoric peoples knew how to count and do basic math.
