I agree with coeurde1ion. There are too many really good movie of different genre that it is not fair to pick ONE favourite movie as it is hard to compare what is best about them.
SOME of the ones I really like are:
Seven Samurai
Akira
Star Wars (all 6)
The Matix
Shrek (1 and 2 BUT NOT 3)
LOTR trilogy
Waking Ned Devine
The Ten Commandments

Imitation of Life
I like the original, but the 1959 remake is my favorite. It's a very powerful movie, especially for that time period. It deals with issues of race discrimination, wealth, fame, success and finding the real meaning of life, self-fulfillment and true happiness.
It tries to answer the questions people often have about what defines a meaningful life.

There are too many good films in the world to have only one favorite.
That being said, if the question is restricted to a certain time period, say for instance the present year, the best movie so far in 2007 is "The Bourne Ultimatum".
