There are several trains per hour, some nonstop, some with one quick change, and they take from 52 minutes to 1 hr. 21 minutes, costing from 14 to 24 euros.
You can catch trains at Frankfurt airport to Mannheim, then take a 18 minute S-Bahn to Heidelberg.
Or if you're in downtown Frankfurt, take a train from Frankfurt Hbf (Hauptbahnhof, Main train station).
You can look at the schedules posted in the stations and buy tickets from the automats in the train stations, or get advice in English at the ticket windows.
You can look up schedules and prices for your exact times and dates at http://www.bahn.de

There are shuttle busses operated by private companies. Sorry, but I can't tell you any web sites because I never used them. I prefer taking the train because I like being rather independent. For connections and fares look at:

There are two train stations in Frankfurt Airport, Fernbahnhof for long distance trains and Regionalbahnhof for local trains (directly beneath Terminal B, which is the main terminal).

I went there once... I flew in from Zagreb and then a driver picked me up and we went by car... There`s a highway, it should take you like an hour or so. I think there were also buses, and public transportation is ok in Germany. I suppose you could always hire a cab if you want to.
Hope this helped:)

As already said, train. It takes roughly an hour, whether it is the more expensive ICE or the less expensive CE - direct connection. Trains are very regular, you can check of course the (english version of) the website www.bahn.de.
