You don't have to get rid of the 12. Put the equation in the form y = (3/2)x - 6. In this form (y = mx + b), the slope is the coefficient of x, and the y-intercept is the constant,
-6.

Why would you want to get rid of the 12? It has nothing to do with the slope. -6 is the y-intercept.

dy/dx=slope=3/2 this is the way we calculate slope by calculus though explaining the reason will be much complicated.
(0,-6) the y intersect of the line.
and (4,0) the x intersect of the line.
slope=(y2-y1)/(x2-x1)
slope={0-(-6)}/{4-0}=3/2
the reason we are calculating the intersects is a line and the coordinate plane makes a triangle and the rise and run would be obvious. since the ratio between rise and run is slope we can formulate it by using the above formula.
so we are using that 12 to find the intersects actualy you are not getting rid of it.
when you know two points in a line you can calculate its slope because this two points also can create a right triangle(by drawing paralels to the axises) which is similar with the one you can create by using coordinate planes. the ratio of the sides would be the slope
