To start with, what kind of check is it? If it's a payroll check, the bank you use will be more than happy to guide you on what to do. Some personal checks from other parties might be slightly more complicated, but to save the hassel, simply go to the bank the check is drawn off of, assuming that it's local. Refund checks can also be taken care of at your bank. Make sure you have a picture id with you, endorse the back of the check on the side your name is written on, usually on the left side of the check. The reason for this is that some banks are a little weird and require that you sign it that way or they will return it for invalid signature. Good luck!!

If you want to get money out of your Bank of America account, you can give the teller a check made out to "Cash"
If someone has given you a check drawn on their account, with your name is the "payee" field, you sign the back of the check and present it to the teller. Depending on the amount of the check and your "standing" they sometimes limit the amount that you can cash.

I disagree with bostonianinmo on a lot, but his answer is the best and most straight forward answer possible to this question.
Take the check to a bank and tell them what you want to do. The do this many times every hour.

take the check to the bank, with ID, sign the back of the check when you get there with your account number on it. stand in line. give it to the teller when they call you. they will give you money back.
or you can deposit some of the check in your account and they will give you the balance as cash.
or you can use the ATM to deposit the check into your account and use money as you need it with your debit card.
the ATM is faster.

go the bofa - and say I want to cash this check. If you have more money than that check you want cashed - you can cash it. If you don't, you have to deposit it then, after 3 days you can cash it.
