I had to replace a car key that my parking lot company lost. I've called them and send two letters, all with no response. Is that enough to take them to court? And how do I get started on that?

Not worth your time unless you are only making minimum wage and have plenty of time to waste. You cannot recoup your time in small claims, only for actual damages plus court costs (process server, etc). Let's say the key cost $100 and court costs are $50. You can sue for $150. How much do you make in a day? (which is what you are going to give up). Unless you make less than $18 an hour, it is not worth it.

Don't use the parking lot company again. Move on.

Yes. You had clear damages caused by them. You need to go to the small claims court where you live, file the paperwork there, pay a fee, and set a court date. Include the court fee in your damages/

Small claims have to be at least three hundred dollars in value. So it just depends how much that key was.
