Well, the lawn should be a little grown out, enough for you to see the weeds. If you mow the lawn, you cut the weed at the stalk and the spray won't get on the foliage (leaves). So, it won't be as effective.
You might also try a weed-and-feed application. It's granules that contain weed killer and fertilizer. You spread it onto the lawn according to the directions and water it in. That way, it doesn't matter when you apply it (before or after mowing).

After reading all of the above answers I will give you my own opinion. You should mow the lawn first to expose the weeds at a low level. The freshly cut leaves on the weeds will be more prone to the effects of the weed killer and the spray will not have to kill off so much foliage before it can attack the root. Be sure to read the instructions for application especially regarding watering after application.
