I personally think the discovery that there are literally billions of other galaxies in the universe, as was shown by the now famous Hubble telescope deep field image. While there have been many past discoveries that altered the course of society much more than this, I can say personally that I was completely blown away by that image, and it changed my entire concept of life, reality, and the nature of the universe.

But seriously, if you refer as in history up to today, then there are many important discoveries. Fire, Wheel, Erath is a planet, Solar System, Galaxy, Universe, E=MC2, Relativity, Gravity, Quantum Mechanics… too many important things.
But if you refer to “of all times” meaning also the future, then forget about it, nobody has come with the greatest discovery of all: why are we here. Either b y chance or with a purpose, but why. When someone discovers that with real proof and is a final answer, then it will be the greatest of all discoveries.

I presume it depends on when you ask this question. All of the answers are correct and you could argue that non of them are correct because the greatest discovery has not been made.
It would also depend on who is asking the question, for example one person might percieve that transferring a humans brain contents into a computer would be the greatest discovery of all time (not yet invented), others might say that fire is the greatest.
Today for example Nasa announce that water vapour has been discovered in a galaxy - this effectively opens the debate on planets forming there that could sustain life - its a tough question.

depends on definition of science.
I like the previous answer of fire or combustion. but it wasn't a researched and planned scientific discovery.
Tanning leather probably falls into the same category.
as for more 'scientific' discoveries...
refining ore into bronze and/or copper was huge in human terms development.
recognizing that disease ws caused by microbial action and not evil spirits would have to be high on the list too...
my opinion anyway

I don't know if you would consider it a discovery but the invention of concrete has help advance transportation, trade and development of our communities. It is used in city water systems, waste systems and is vital to all countries infrastructures.

Germ Theory which gave rise to vaccines for the immune system where you can give a weakened virus to teach the immune system to respond faster the natural virus.
Followed by genetic theory and the structure of DNA that confirms it.
From physics: E=mc^2 would have to have the furthest reaching theory.

I could not say that there is ONLY one great discovery.
fire
wheel
food presevation
shaping metals
germs
computers
black holes
string theory
I'd say
DNA
OR
Epigenetics
(how chemicals of any sort (food, pollution or hormones) can permanently effect our DNA and the DNA of our children)
we have a little bit more work, but soon creating superhumans won't be a problem.
I'm only concerned about WHO will own this new life
