If your mother is the subject of the sentence, then write it like this:
"My mother and I were inseparable."
However, since "no one" is the subject of this sentence, then write it like this:
"No one could ever come between my mother and me."
The difference is that "mother and I" is the object of the sentence, not the subject.

would you say "between we" or "between us"? Between is a preposition and always takes an objective pronoun after it. "Us" is an objective pronoun. Usually toward the front of the sentence you use nominative pronouns and toward the end objective ones. Look them up.
