Depends with the situation and depends with the gender.

You know, when standing in line at a cash register, if someone tries to cut in, I tell them the line ends in the back. But if I see somebody right behind me with just 2 or 3 things, I tell them to go before me. strangely enough, I've met a number of people who feel the same way about me.

Generally, I will not allow it and ask the person to take his rightful slot in the line. But I have found myself grudgingly allowing someone to go ahead - - - in airport immigration counters when the person pleads that he is about to miss his flight (and there is no time to lecture this person about coming to the airport allowing for traffic, etc). I would not want it in my conscience if the poor OFW misses employment opportunities just because I asserted my right. I just shake my head.

I will just tap them at the shoulder and point the back end of the line without saying a word.

It is contextual for me.
I will let the person go if I am not in hurry whereas he may need to rush as it happens at airport gates.
If I have been standing for long and tired, I will protest.
I have come to accept these things in this busy mad world.

Sometimes my blood is boiled, but I always try to maintain a good composure because I know those types of rude people are not worthed for me to get into a trouble with the law!

I usually pat the person's shoulder and say "excuse me, the end of the line is over *there*". They usually go straight to the back of the line if I throw in my evil stare. >O_O<

I say "excuse me, but I'm in line" and then point toward the end of the line.

I get ballistics, of course --- esp. if it's the bathroom line. Food line, grocery line, concert line, movie line, airport line, etc., I stay calm but I give them THE LOOK.
