You need to research and see where you are spending the most money. Ex: payroll, supplies, reducing markdowns etc. every business is different and you need to focus on making a difference where you can.

Change all light bulbs to energy saving florescent, if possible look for govt green rebate where available. Hire students or housewife do part-time replace those full timers with benefit and eliminate over-time. Your store will see more work forces but majority part-timers who save you from paying health care, unemployment insurances and other expenses. Re-evaluate inventories from supplier, shop around for better bargain or direct import buying bulk and pass on the saving to consumer which will generate more sells improve store image.

Your energy is better spent on "how do I increase customer spending"...after all you are in the "Making Customers" business.
The answer lies in providing better than exceptional customer service, and increased efforts on creative marketing, and an ample advertsing budget.
If you start down the road of cutting costs you run the rick of cutting too much....the next thing you know, you begin to hamper your ability to function at a quality level.

Buy your supplies from a Club such as Sam's Club or Costco. Also, if you have a Dollar General in your area, they are comparable also. Cut overhead and overtime!

You did not say what kind of store . . .
Different business have different costs .
So , the only option is staff reduction on a general basis .
