turn off the water supply to the toilet. little faucet knob in back of or under toilet tank, coming out of floor or wall with small hose attached.
flush the toilet. when water is gone, get a wire from coathanger or something, or either pliers and see if you can remove the obstruction.

Take the loo out, or get a plumber to do it, if you push the block past the u-bend, it may get stuck in the drains underground, then you will have to dig it out! Toilet rim blocks are the worst idea ever, especially if you have children, they should be banned! (the loo blocks, not the kids, lol), I have removed lots of these over the years!

The toilet might have to be removed to get at the blockage ( if went past the Ubend). You can either buy one or rent one from plumbing supply stores ( i think home improvement stores might too) called a plumbing snake - its a large metal coil that feeds down into the piping and either cleans out/extracts objects.
Id suggest not using the block thingy again ;)

If you can see it, and the water is clean, just grab it with your hand. You can wash your hand afterwards. Or try to hook it with a piece of bent wire, like a coat hangar.
If you can't see it, and you have another toilet, just be sure that there's water in the toilet. It will soften and dissolve the block after a day or two, enough that you can flush it or retrieve it with the wire.
