I'd first redo the rough plumbing for the tub and toilet, where you want them. Then lay 3/4" ply across the joists. Use screws to fasten it down well. Drill out for the drain area for the tub and toilet, install tub ON the new subfloor.
Put a strip of tar paper along the edge where it sits on the floor.

- Remodeled many a bathroom.

BTW, what the hell is pol talking about?

I want so much to assume that any decent, original contractor would have installed a complete sub floor first; knowing the locations of the plumbing, and the size of the area for a tub or shower stall. IF that tub is on a second floor, OR if it happens to be above a basement or crawl space. If that isn't the case; I as well as others suggest you do install a sub floor taking into account the holes to be drilled for the plumbing.

Obviously there may have been reasons initially? even if only for more clear access underneath?

Sometimes SHORTCUTS are acceptable, even for contractors, but I can't imagine a need to leave a tub installed just on floor rafters. That said however,,, the exception might be; if you're on a "slab" in that case it is a waste. Your sub and finish floor would be installed on the same surface, and would be finished to BUTT the tub AFTER it was installed.

If the base of the bath is attractive it can make a bathroom appear bigger to continue same flooring throughout including under the bath rather than boxing the bath in.

Only because it should have been done to begin with and it adds structural integrity. Avoid areas where plumbing to the trap has to be done.
