Straight from the SNL website...

Q: How can I become a cast member on the show?

A: "SNL" does not hold open auditions or accept audition tapes. Unfortunately, you can't go to them; they have to find you. Most of their performers are culled from the comedy groups The Groundlings and Second City, in Los Angeles and Chicago, respectively. If they see a performance and are impressed with a particular performer, they invite the person for an "SNL" audition.

"As an avid fan of SNL I would have to say move to Chicago, somehow join Second City and show Lorne your unique comedy . Know many impersonations and make them laugh. This was my audition Lorne. Thank you for the great comedy. And tonight From New York it's Saturday Night."

Usually, the person is approached by a representative from the show.

So if you want to be a cast member, I'd suggest start with creating a buzz about yourself, and then backing it up, and hope someone brings it to Lorne's attention.

Most of the people on SNL come from regional improvisational comedy troupes like Second City in Chicago and are seen performing there and invited to join SNL.

they usually get chosen from comedy troupes across the Country and Canada, such as second city, the groundlings, etc.
so you have to already be doing comedy, and get a recommendation.
