Do you NEED? It depends on what you have. If you have an Ethernet cable and two cards, use it, as it is the fastest. If you have a USB cable but no Ethernet, and both computers have USB, then use USB. If you have a CD- burner, but no cables, use that. If you have a Flash drive, use that.
Personally? I mostly use an Ethernet cable, although for some computers I use a Flash drive.

Don't use USB for this! Although some USB cables have the conncetors that allow you to plug it in, it may not be a cable desinged for this and it can smoke one or both computers.
Go with Ethernet. If you don't have a router (or hub or switch) then use a "half twist" ethernet cable....or an ethernet card with "autosense".

Ethernet cable would be easier and cheaper, especially if both systems already have the network cards
BTW, you do not use the regular USB cable to do the job. just to move your files. Something like Laplink's :

http://www.laplink.com/etc (for Vista)
Buy it only if you plan to regularly transfer data between two computers. And if you have only USB1 , it will be pretty slow.
So, use Enthernet cables, if you can - doesn't cost you much and probably less of a hassle.

It depends on how you want to do it.
The most common way is Ethernet cable.
If the cable is not there already and you plan to data transfer not very frequently then Flash drive will be the best thing i would do.
